Wilson began her career as Toula in the SBS comedy series Pizza (2003–2007, 2019) and later in the sketch comedy series The Wedge (2006–2007) after graduating from the Australian Theatre for Young People in 2003.
In the musical comedy series Bogan Pride, she wrote, produced, and starred (2008). Wilson starred in the comedies Bridesmaids and A Few Best Men, both released in 2011, shortly after moving to the United States.Wilson was named one of Variety’s “Top Ten Comics to Watch for 2013” after appearing in the comedies What to Expect When You’re Expecting, Struck by Lightning, and Bachelorette in 2012.

Wilson received A$4,500,000 in damages on September 13, 2017. “Today’s verdict is a significant record—roughly it’s four times the highest previous verdict in a defamation case in Australia,” Wilson’s lawyer, Richard Leder, said. “Any dollars I receive will go to charity, scholarships, or invested in the Australian film industry to provide jobs,” Wilson said in a tweet in June 2017. The damages amount was challenged by Bauer Media.
Bauer Media won its appeal on June 14, 2018, and the damages were reduced to $600,000, requiring her to repay A$4.1 million plus interest of A$60,000. She was also responsible for paying for 80% of Bauer’s appeal costs. Wilson filed an appeal with the High Court on July 11, 2018, after announcing her intention to appeal the decision. On November 16, 2018, the High Court dismissed the petition.
